ShareonEmailFacebookTwitterLinkedInRedditWechatAbstractAxonalregenerationintheperipheralnervoussystemisgreatlysupportedbySchwanncells(SCs).Afternerveinjury,SCsdedifferentiatetoaprogenitor‐likestateandefficientlyguideaxonstotheiroriginaltargettissues.ContactandsolublefactorsparticipateinthecrosstalkbetweenSCsandaxonsduringaxonalregeneration.HereweshowthatdedifferentiatedSCssecretenano‐vesiclesknownasexosomeswhicharespecificallyinternalizedbyaxons.Surprisingly,SC‐derivedexosomesmarkedlyincreaseaxonalregenerationinvitroandenhanceregenerationaftersciaticnerveinjuryinvivo.Exosomesshiftthegrowthconemorphologytoapro‐regeneratingphenotypeanddecreasetheactivityoftheGTPaseRhoA,involvedingrowthconecollapseandaxonretraction.Altogether,ourworkidentifiesanovelmechanismbywhichSCscommunicatewithneighboringaxonsduringregenerativeprocesses.WeproposethatSCexosomesrepresentanimportantmechanismbywhichthesecellslocallysupportaxonalmaintenanceandregenerationafternervedamage.GLIA2013;61:1795–1806
